Development and validation of a microfluidic reactor for biofilm monitoring via optical methods

چکیده
We present the design, fabrication, and verification of a microfluidic platform for optical monitoring of bacterial biofilms. Biofilm formation characterizes the majority of infections caused by bacteria that are developing increased resistance to traditional antibiotic treatment, necessitating the development of reliable tools not only for study of biofilm growth, but also for in situ examination of the response to applied stimuli. The presented platform was used to continuously and non-invasively observe the dependence of Escherichia coli biofilm formation on bacterial signaling by monitoring the change in biofilm optical density over the growth period. Results were corroborated by measurement of biofilm morphological properties via confocal microscopy, and statistical analysis was applied to verify the repeatability of observed optical and morphological differences in the biofilms formed. The presented platform will be used to characterize biofilm formation and response in drug discovery applications. (Some figures in this article are in colour only in the electronic version)

Macrostructural Biofilm Characterization via Textural Image Analysis by SGLDM and GLRLM
The macrostructure development of biofilms grown in an annular reactor and in a rotating biological contactor has been monitored using the Spatial Gray-level Dependence Method (SGLDM) and the Gray-Level Run Length Method (GLRLM). Due to the large number of descriptors, Principal Components Analysis was used to sort the results. Both methods look promising for biofilm monitoring.

A novel microfluidic reactor for biofilm growth and in situ characterization using time-of-flight secondary ion mass spectrometry (ToF-SIMS) was constructed to enable two-dimensional chemical imaging of hydrated biofilms. We demonstrate the detection of characteristic fatty acid fragments from microfluidic reactor-grown biofilms and illustrate advantages of hydrated-state ToF-SIMS imaging.

The purpose of this study is to investigate the accuracy of predictions of aniline removal efficiency in a moving bed biofilm reactor (MBBR) by various methods, namely by RBF, ANFIS, and fuzzy regression analysis. The reactor was operated in an aerobic batch and was filled by light expanded clay aggregate (LECA) as a carrier for the treatment of Aniline synthetic wastewater. Exploratory data an...
